WW domain binding protein 1-like (WBP1L, also known as OPAL1 or outcome predictor of acute leukaemia 1) is a transmembrane adaptor protein expressed in a broad range of hematopoietic and non-hematopoietic cells[1][3]. It contains several WW domain binding motifs and interacts with multiple NEDD4-family E3 ubiquitin ligases (including WWP2, ITCH, WWP1, and NEDD4L)[1]. WBP1L regulates the ubiquitination and cell-surface expression of the chemokine receptor CXCR4, a critical modulator of hematopoiesis and leukaemia cell survival[1][3]. Deficiency in WBP1L results in enhanced CXCR4 signaling and altered B cell development, as well as enhanced efficiency of bone marrow transplantation in mice[1]. WBP1L expression is especially increased in B cell progenitor acute lymphoblastic leukaemia (BCP-ALL) with ETV6-RUNX1 translocation, correlating with a favorable prognosis[1]. There are no reports of direct drug-targeting or safety concerns related to WBP1L, but it is a useful biomarker for prognosis in acute leukaemia[1][3].
