Overview

WW domain binding protein VOPP1 (VOPP1) is a protein coding gene implicated in cancer, most notably glioblastoma and breast carcinoma[1][6]. VOPP1 increases the transcriptional activity of the nuclear factor kappa B subunit 1 (NFKB1) by facilitating its nuclear translocation, DNA-binding, and associated apoptotic response, especially when overexpressed[4][5][7]. It is proposed to sequester the tumor suppressor WWOX in lysosomal vesicles, thereby regulating WWOX’s tumor-suppressive activity[1]. Subcellularly, VOPP1 localizes to the cytoplasmic vesicle membrane, late endosome, and lysosome[3]. There is not a direct small-molecule or biologic targeting VOPP1 in clinical use, but its involvement in NFKB signaling and oncogenesis makes it a potential target of interest for drug discovery in cancer therapy[6].
