Overview

WW domain-containing oxidoreductase (WWOX) is a tumor suppressor protein that plays a critical role in apoptosis, development, and cellular stress responses. It contains two N-terminal WW domains and a C-terminal short-chain dehydrogenase/reductase (SDR) domain. WWOX interacts with numerous proteins via its WW domains, influencing various cellular pathways. Loss of WWOX expression is frequently observed in many cancers, suggesting its importance in maintaining genomic stability and preventing tumor development.

Mechanism of action

WWOX acts as a tumor suppressor by inducing apoptosis and inhibiting tumor growth. It functions as an adaptor protein via its WW domains, interacting with various signaling proteins containing PPxY motifs. The SDR domain is involved in redox reactions, particularly with steroid substrates.
