Overview

WW domain-containing transcription regulator 1 (TAZ) is a transcriptional coactivator and downstream regulatory target within the Hippo signaling pathway, crucial for controlling organ size and suppressing tumors by restricting cell proliferation and promoting apoptosis. It is structurally related to Yes-associated protein (YAP) and contains both a WW domain for interaction with PPXY motifs in transcription factors and a C-terminal PDZ-binding motif, which enables subcellular localization and function. TAZ activity is negatively regulated by 14-3-3 protein-mediated nuclear export and positively regulated via PDZ domain interactions. It modulates the transcriptional activity of multiple factors and has important roles in stem cell self-renewal, cell proliferation, epithelial-mesenchymal transition, and differentiation, with its dysregulation implicated in cancer development and progression[1][3][4][5][6].
