Overview

WW domain-containing transcription regulator 1 antisense RNA 1 (WWTR1-AS1) is a long non-coding RNA (lncRNA) located at chromosome 3q25.1, transcribed antisense to the WWTR1 gene. It is significantly overexpressed in multiple cancers, including breast cancer, head and neck squamous cell carcinoma, and cervical squamous cell carcinoma, where its expression correlates with higher tumor grade, metastasis, and worse prognosis. Functionally, WWTR1-AS1 promotes cancer cell proliferation, migration, invasion, stemness, and resistance to apoptosis, possibly via modulating the stability and expression of its sense partner WWTR1 mRNA and regulating transcriptional and signaling networks. Elevated WWTR1-AS1 can serve as a reliable diagnostic and prognostic biomarker in several cancer types, and preclinical studies suggest it may be a candidate therapeutic target, though no direct drugs are available and clinical targeting remains investigational.
