Overview

X antigen family member 1B (XAGE1B) is a member of the cancer-testis (CT) antigen family, specifically the XAGE subfamily, within the broader GAGE gene family[5][4]. It encodes a nuclear protein, produced predominantly in the testis and various tumor types—notably lung adenocarcinoma, Ewing's sarcoma, rhabdomyosarcoma, and hepatocellular carcinoma[6][4][5]. There are multiple transcript variants of the XAGE-1 gene (a, b, c, d, e), but XAGE-1b is the dominantly expressed cancer-associated variant[2][6]. The protein is highly immunogenic in cancer patients and contains a nuclear localization signal[6][4]. Expression in normal tissue is highly restricted to the testis; it is otherwise nearly undetectable outside of tumors[6][4]. Functional ly, the gene is annotated for protein binding, with potential involvement in cell signaling and complex formation, though its precise molecular role remains poorly defined[3][6]. XAGE1B is studied as both a possible diagnostic and prognostic biomarker in cancer—particularly for hepatocellular carcinoma, where serum XAGE-1b mRNA has been shown to have high sensitivity and specificity[2]. Its expression is linked with aggressive clinical features and disease recurrence in this context. No specific drugs targeting XAGE1B are currently described, but as a cancer-testis antigen, it is a potential target for cancer immunotherapy, such as cancer vaccines or adoptive T-cell therapies[6][2][5]. The restricted tissue distribution suggests a potentially favorable therapeutic window, but the functional effects and safety of XAGE1B-directed interventions are not yet well defined.
