Overview

X antigen family member 1B (XAGE-1b) is a prominent cancer/testis antigen (CTA) belonging to the GAGE family, primarily expressed in the testis and placenta but aberrantly overexpressed in various malignancies, most notably lung adenocarcinoma (UniProt Q9HD64; GeneCards). It is a nuclear protein that likely functions as a transcription factor, promoting cell proliferation and metastasis while inhibiting apoptosis in cancer cells (PubMed: 25963468). Due to its high immunogenicity and restricted expression in normal tissues, XAGE-1b is a major target for cancer immunotherapy, including peptide-based vaccines and adoptive T-cell therapies (PubMed: 16061866). Clinical studies have demonstrated that XAGE-1b can induce both humoral and cellular immune responses in patients, making it a valuable biomarker for prognosis and a candidate for targeted treatment (PubMed: 26936399). Therapeutic approaches often utilize synthetic long peptides or dendritic cells pulsed with the antigen to stimulate cytotoxic T-lymphocyte activity against tumors (PubMed: 25963468). The restricted expression profile of XAGE-1b minimizes the risk of off-target toxicity, although local injection site reactions are common in vaccine trials (PubMed: 1.1.5).
