Overview

X antigen family member 2 (XAGE2) is a member of the XAGE subfamily within the GAGE cancer-testis antigen family. XAGE2 is primarily expressed in normal testicular tissue but is also found in a range of tumors, including Ewing’s sarcoma, rhabdomyosarcoma, breast cancer, and germ cell tumors[1][3][6]. It is part of a broader class of cancer-testis (CT) antigens, which are largely restricted to germ cells in the testis and certain malignancies, making them attractive candidates for cancer immunotherapy and as potential biomarkers. Like related GAGE/XAGE antigens, its main functions are believed to involve immune recognition and a possible role in supporting tumor cell proliferation and survival, though the exact cellular mechanisms for XAGE2 remain poorly described[1][4][6]. There are currently no approved drugs targeting XAGE2 directly, but it is being considered for vaccine and immunotherapy development due to its cancer-restricted expression profile[6].
