Overview

X antigen family member 3 (XAGE3) is a protein coding member of the GAGE family, classified as a cancer/testis (CT) antigen, meaning it is typically restricted in expression to human germ cells (testis and placenta) and a broad range of cancers[3][2]. XAGE3 is absent or expressed at very low levels in most somatic tissues and upregulated in various malignancies, making it a promising immunotherapy target and biomarker[2][4][10]. The biological function of XAGE3 remains incompletely characterized; however, GAGE family CT antigens commonly regulate cell proliferation, inhibit apoptosis, and contribute to cancer cell survival and metastatic potential[4]. XAGE3 has two known transcript variants and shares sequence similarity with other GAGE/PAGE proteins[3]. Its expression may be induced in cancer cell lines by demethylating agents (e.g., decitabine), a property used to enhance antigen presentation for immunotherapy[6]. XAGE3 is not currently associated with any approved drugs or targeted agents, but is under investigation as a tumoral marker for immunotherapeutic strategies and prognosis in cancer[8][10].
