Overview

X antigen family member 5 (XAGE5) is a protein-coding gene belonging to the XAGE (X Antigen Family) subfamily, which is part of the GAGE family of cancer/testis antigens (CTAs)[9]. This protein is primarily expressed in reproductive tissues (testis, placenta) and a broad array of malignancies, where its presence is associated with tumorigenesis and poor clinical outcomes in several cancers[1][9]. XAGE5 shows sequence similarity with other GAGE/PAGE proteins, and although its function remains to be fully elucidated, it may be involved in cellular processes supporting proliferation and resistance to cell death in cancer cells, as well as maintenance of pluripotency in germ cells[1][7]. Its immunogenic properties and restricted expression profile make it a candidate biomarker and therapeutic target in immuno-oncology, particularly in vaccine development for cancers such as sarcoma and lung cancer[1][9]. At present, no drugs are known to specifically target XAGE5, but it represents a promising avenue for future antitumor immunotherapies[1][9].
