Overview

Y-box-binding protein 3 (YBX3), also known as ZONAB, DbpA, and CSDA, is a multifunctional cold shock domain-containing protein that binds DNA and RNA, acting primarily as a transcriptional and post-transcriptional regulator. In epithelial cells, YBX3 dynamically shuttles between tight junctions and the nucleus, where it regulates genes critical for cell cycle progression, cellular differentiation, and responses to environmental stress. YBX3 binds mRNA (including 3′-UTRs of key transcripts) to stabilize and enhance their translation, influencing proliferation and apoptosis. Dysregulation of YBX3 is implicated in the pathogenesis and progression of multiple malignancies, where its overexpression is linked to enhanced proliferation, invasion, and chemoresistance. Recent studies demonstrate roles in tumor immune evasion, regulation of epithelial–mesenchymal transition, and potential antiviral activity against influenza A virus
