Overview

Y-chromosome bearing bovine spermatozoa are the male gametes in cattle that carry the Y-sex chromosome, which dictates the development of male offspring. These cells are not therapeutic targets in the traditional medical sense but are critical targets for selection in agricultural biotechnology to manage herd sex ratios (Garner, D. L., & Seidel, G. E., 2008). The primary method for identifying and isolating these cells is flow cytometric sorting, which exploits the approximately 3.8% difference in DNA content between X- and Y-bearing bovine sperm (Johnson, L. A., & Welch, G. R., 1999). During this process, sperm are typically treated with the fluorescent dye Hoechst 33342, which binds to DNA and allows for quantification via laser excitation. While this technology is widely used in the dairy industry to produce female replacements, the sorting process can cause physiological stress to the sperm, including membrane damage and reduced motility (DeJarnette, J. M., et al., 2009). Consequently, sex-sorted semen often exhibits lower conception rates compared to conventional unsorted semen. There are no known drugs that target these cells for disease treatment, as their relevance is strictly limited to reproductive management and genetic selection.
