YEATS domain-containing protein 2 (YEATS2) is an evolutionarily conserved chromatin reader and scaffold protein that serves as a core component of the ATAC (Ada Two A Containing) complex, which has acetyltransferase activity targeting histones H3 and H4. YEATS2 harbors a YEATS domain that specifically recognizes various lysine acylation marks on histones, including crotonylation, benzoylation, and acetylation, positioning the ATAC complex to regulate gene expression in response to chromatin signals. YEATS2 thereby orchestrates transcriptional co-regulation and chromatin remodeling. Aberrant YEATS2 function, domain mutations, or amplification have been implicated in cancer and certain epilepsy syndromes. YEATS2 is considered a rational epigenetic drug target, with several selective chemical inhibitors and PROTACs advancing in research pipelines to disrupt oncogene-driven or pathological gene expression programs.
Inhibition of YEATS2 restricts its ability to bind acylated lysines on histones, thereby disrupting chromatin remodeling and transcriptional activity driven by the ATAC complex in disease states PROTACs (proteolysis-targeting chimeras) directed against YEATS2 and related domains are under active research for targeted protein degradation
