Overview

The Yellow Fever 17D (YF-17D) vaccine antigens represent a complex set of viral proteins that stimulate the human adaptive immune system to provide long-term immunity against the yellow fever virus (Pulendran, B., Nature Reviews Immunology, 2009). The vaccine is a live-attenuated virus that infects dendritic cells, activating multiple Toll-like receptors (TLR2, 7, 8, and 9) to induce a potent innate immune signal (Querec, T., et al., Nature Medicine, 2009). This innate activation leads to a robust adaptive response, including the production of high-titer neutralizing antibodies and the generation of polyfunctional CD8+ T cells (Akondy, R. S., et al., Nature, 2009). YF-17D is considered one of the most effective vaccines ever developed, often providing lifelong protection with a single dose (WHO, 2023). However, it is associated with rare but serious adverse events such as viscerotropic and neurotropic diseases, particularly in specific vulnerable populations (CDC, 2023).

Mechanism of action

Activation of the innate immune system via Toll-like receptors (TLR2, 7, 8, 9) on dendritic cells, which coordinates a broad adaptive immune response involving neutralizing antibodies and CD8+ T cells (Querec et al., 2006; Pulendran, 2009).
