Overview

The YES-associated protein–transcriptional enhancer factor domain complex (YAP–TEAD) is a key transcriptional regulator involved in cell proliferation, growth, and survival. YAP, a transcriptional co-activator, interacts with TEAD transcription factors to promote the expression of genes involved in these processes. The activity of YAP is regulated by the Hippo signaling pathway, with phosphorylation leading to cytoplasmic retention and degradation, while dephosphorylation allows nuclear translocation and TEAD binding. Dysregulation of the YAP-TEAD complex is implicated in various cancers, making it a potential therapeutic target.
