Overview

Yes-associated protein 1 (YAP1) is a transcriptional coactivator and a key downstream effector of the Hippo signaling pathway. It plays a central role in regulating organ size, tissue regeneration, cell proliferation, apoptosis suppression, and tumorigenesis. YAP1 promotes the expression of genes involved in cell proliferation and suppresses apoptotic genes. In its dephosphorylated state, it translocates to the nucleus where it binds TEAD family transcription factors to drive target gene expression. When phosphorylated by upstream Hippo pathway kinases such as LATS/MST, it is sequestered in the cytoplasm or targeted for degradation.
