YAP1 mRNA is the messenger RNA that encodes Yes-associated protein 1, a key transcriptional coregulator within the Hippo signaling pathway. YAP1 protein operates mainly by binding to TEAD family transcription factors, driving expression of genes involved in cell proliferation, suppression of apoptosis, and organ size regulation. Dysregulation and amplification of YAP1 contribute to multiple human cancers, making it an established oncogenic driver. While YAP1 protein is a major therapeutic target, especially in oncology, the mRNA itself may be targeted directly by antisense or RNAi strategies; however, the canonical context is the protein. Interfering with YAP1 signaling is being pursued by direct YAP1-TEAD inhibition and related mechanisms, but such interventions must balance anti-tumor efficacy with potential impacts on normal tissue growth and repair.
