Overview

YTH domain-containing protein 1 (YTHDC1) is a nuclear RNA-binding protein encoded by the human *YTHDC1* gene. It plays a critical role in regulating alternative splice site selection during pre-mRNA processing, localizing to dynamic subnuclear compartments known as "YT bodies" that appear during S-phase and disperse in mitosis. The defining feature of this family is the highly conserved YTH domain, which binds single-stranded RNA motifs—especially those containing N6-methyladenosine (m6A) modifications—thereby influencing exon inclusion or exclusion in mature transcripts. Through its control over alternative splicing, particularly of genes involved in tumorigenesis such as VEGF, BRCA1, and progesterone receptor, it has been implicated in cancer biology. In prostate cancer specifically, it interacts with metadherin to modulate tumor-related gene splicing. The broader family of YTH proteins are found only in eukaryotes and are key regulators of mRNA fate via recognition of m6A marks on RNAs[1][3][4].
