YTH N6-methyladenosine RNA binding protein 2 (YTHDF2)

Target
YTHDF2
Molecular classification
RNA binding protein, m6A reader, Post-transcriptional regulator
01

Overview

YTH N6-methyladenosine RNA binding protein 2 (YTHDF2) is an RNA binding protein that recognizes and binds N6-methyladenosine (m6A) modifications in RNA. It acts as a key "reader" of the m6A epitranscriptomic mark, primarily in the cytoplasm. Upon binding m6A-modified mRNAs, YTHDF2 recruits the CCR4—NOT deadenylase complex to promote rapid degradation of these transcripts, thus regulating mRNA stability and turnover. This post-transcriptional control impacts cell proliferation, apoptosis, and other essential biological processes. Dysregulation of YTHDF2 is implicated in cancer progression (notably bladder cancer) and may serve as a prognostic biomarker.

Mechanism of action

Compounds or molecules targeting YTHDF2 would be expected to modulate mRNA stability by altering recognition or degradation of m6A-marked mRNAs

03

Biological functions

mRNA stability regulation (promotes degradation of m6A-containing mRNAs)RNA metabolism, including pre-mRNA splicing, export, and translationRegulation of cell proliferation, apoptosis, cell cycle, viability, migration, and invasion
04

Disease associations

Cancer (notably associated with bladder cancer and other malignancies)Potential roles in polycystic ovary syndrome and mitochondrial dysfunction
05

Safety considerations

Potential for broad effects on mRNA metabolism, leading to possible off-target disturbances in cell functionModulation may impact cell proliferation and apoptosis, affecting tumorigenesis or other cellular homeostasis
06

Biomarkers

Proposed as a diagnostic/prognostic biomarker in cancer, particularly bladder cancer
