VP35 is a multifunctional protein encoded by the Zaire ebolavirus. It is essential for viral replication, pathogenesis, and immune evasion. VP35 acts as an essential cofactor of the viral RNA polymerase complex, facilitating both transcription and replication of the viral genome. A key function of VP35 is to antagonize host innate immunity by inhibiting type I interferon (IFN) production. The C-terminal domain binds double-stranded RNA (dsRNA), blocking recognition by host sensors such as RIG-I and MDA5. VP35 also suppresses dendritic cell maturation, further dampening immune responses during infection. Due to its central roles in both viral life cycle progression and immune evasion, VP35 represents an attractive target for antiviral drug development.
