Overview

Zaire ebolavirus Viral Protein 35 (VP35) mRNA is the genetic transcript that encodes the VP35 protein, a critical multifunctional component of the Ebola virus. The VP35 protein acts as an essential non-catalytic cofactor for the viral RNA-dependent RNA polymerase (L), which is required for the replication and transcription of the viral genome (PMID: 25552711). Furthermore, VP35 is a primary factor in the virus's ability to evade the host immune system by inhibiting the production of Type I interferons and blocking RIG-I-like receptor signaling (PMID: 19114480). By targeting the VP35 mRNA, therapeutic interventions aim to halt viral replication and restore the host's innate immune response. Drugs such as small interfering RNAs (siRNAs) and phosphorodiamidate morpholino oligomers (PMOs) have been designed to bind specifically to this mRNA sequence to induce its degradation or block its translation (PMID: 24086301). Clinical candidates like TKM-Ebola and AVI-7537 have demonstrated the potential of this target in treating Ebola Virus Disease by significantly reducing viral titers in experimental models (PMID: 26119835).
