ZBTB40 intronic transcript 1 (ZBTB40-IT1) is a long non-coding RNA gene located within or near the ZBTB40 locus, which is associated with bone density and osteoporosis susceptibility. ZBTB40-IT1 acts by suppressing osteoblast differentiation and promoting osteoclast activity, therefore playing a role in bone metabolism. Its expression is regulated by SNPs linked to osteoporosis risk, and it modulates downstream targets including WNT4, RUNX2, and COL1A1. Currently available evidence supports its functional relevance in osteoporosis but does not indicate that it is a therapeutic target for drugs or small molecules[1][4].
