Zeta-chain-associated protein kinase 70 (ZAP70) is a cytoplasmic tyrosine kinase critical for T cell receptor (TCR) signaling. It transduces signals from activated TCR complexes, driving T cell activation, differentiation, proliferation, and cytokine secretion. ZAP70 contains two N-terminal SH2 domains that bind to phosphorylated ITAMs on the TCR ζ chain, and a C-terminal kinase domain. Mutations in ZAP70 can cause severe combined immunodeficiency (SCID), characterized by the absence or dysfunction of CD8+ T cells.
