Overview

Zika virus nonstructural protein 1 (NS1) is a multifunctional, glycosylated viral protein essential for Zika virus replication and pathogenesis[1][2][3][5]. NS1 forms homodimers that participate in viral replication complexes within the host cell, and is also secreted as a hexameric lipoprotein particle into the extracellular space, where it interacts with components of the host immune system, facilitating immune evasion and contributing to disease processes[2][3][5]. Structurally, NS1 displays conserved regions among flaviviruses but displays diverse electrostatic properties at host-interaction interfaces, suggesting different modes of pathogenesis among related viruses[1][2][3][5]. NS1 serves as the major antigenic marker for Zika virus infection, making it important for diagnostics and a potential therapeutic target, though no specific drugs are currently approved that interact directly with Zika virus NS1[2]. Cross-reactivity with other flaviviruses and its role in immune modulation pose notable challenges for therapeutic and vaccine development.
