Zinc finger AN1-type domain 2A (ZFAND2A)

Target
ZFAND2A
Molecular classification
Zinc finger protein, Heat shock protein (inducible), Stress-response mediator, Other (protein coding gene, not a classic receptor/enzyme/transcription factor)
01

Overview

Zinc finger AN1-type domain 2A (ZFAND2A, AIRAP) is a stress-responsive protein with predicted zinc ion binding activity, primarily localized to the nucleus and endoplasmic reticulum. ZFAND2A expression is tightly regulated by heat shock factor 1 (HSF1) and rapidly induced by heat shock and proteotoxic stressors—including bortezomib—positioning it as a mediator of cellular defense mechanisms against protein misfolding and aggregation. ZFAND2A participates in proteasome-mediated protein catabolism, positive regulation of proteasomal activity, and adapts cell responses to environmental challenges such as arsenic exposure and elevated temperature. It is not considered a classic therapeutic target, but its regulation and activity are relevant in the contexts of cancer therapy and cellular stress adaptation.

Mechanism of action

Drug-induced (bortezomib): Proteasome inhibition leads to proteotoxic stress and activation of ZFAND2A via heat shock factor 1 (HSF1)-mediated transcription

Biological functions

Zinc ion bindingProteasome-mediated ubiquitin-dependent protein catabolic processProtein targeting to endoplasmic reticulumCellular response to heat shock/proteotoxic stressCellular response to arsenic-containing substancesPositive regulation of proteasomal activity during stress
04

Disease associations

Cancer (expression induced by bortezomib, a cancer therapy)Other (roles in stress adaptation may impact cellular homeostasis in disease but direct disease causality is not established in major databases)
05

Safety considerations

None reported in relation to therapeutic targeting—its induction is part of the normal cellular protective response, not known to cause toxicity
06

Interacting drugs

Bortezomib (induction of ZFAND2A expression observed upon treatment)
07

Biomarkers

Potential marker for cellular stress response, especially heat shock and proteasome inhibition in cancer therapy (e.g., bortezomib sensitivity)
