Overview

Zinc finger and BTB domain-containing protein 1 (ZBTB1) is a member of the ZBTB (zinc finger and BTB/POZ domain-containing) family of transcription factors that function primarily as transcriptional repressors.[1][2][3][6] ZBTB1 contains an N-terminal BTB/POZ dimerization domain and multiple C2H2-type zinc finger domains at the C-terminus, enabling it to bind specific DNA sequences and interact with chromatin-modifying complexes.[1][3][5] It is specifically essential for development of the lymphoid lineage, especially T cell development; absence leads to lack of T cells and reduction in B and NK cells, recapitulating a SCID-like immunodeficiency phenotype in mice.[1][4] ZBTB1 is localized to the nucleus and appears to modulate transcription of key genes required for lymphocyte differentiation, possibly via chromatin remodeling and repression of cAMP response element (CRE)-mediated transcription.[1][3][6] Although critical for immune system development, there are no approved drugs or well-characterized small molecule inhibitors that selectively target ZBTB1, and its direct role as a therapeutic target or biomarker has not been described in current literature.[2][3][6]
