Overview

Zinc finger and BTB domain-containing protein 2 (ZBTB2) is a member of the BTB/POZ family of transcription factors that acts primarily as a transcriptional repressor. It contains zinc finger motifs mediating DNA binding and a BTB/POZ domain that facilitates protein-protein interactions. ZBTB2 represses genes within the ARF-HDM2-p53-p21 axis critical for cell cycle control, acting as a master regulator that can inhibit the cell cycle arrest function of p53 by direct DNA binding and interaction with transcription factors such as Sp1. ZBTB2 associates with chromatin-modifying complexes (including NuRD and HiRA) and regulates chromatin structure during stem cell differentiation and development. Dysregulation or abnormal expression of ZBTB2 is implicated in cancer biology as a potential proto-oncogene and is associated with certain inherited retinal degenerative diseases.
