Overview

Zinc finger and BTB domain-containing protein 20 (ZBTB20) is a transcription factor characterized by an N-terminal BTB/POZ domain and multiple C2H2-type zinc finger motifs at the C-terminus[1][3][5]. ZBTB20 acts mainly as a transcriptional repressor, regulating diverse biological processes such as cellular development and differentiation, metabolism, and innate and adaptive immune responses[1][2][6]. It is important in hematopoietic tissues and is highly homologous to BCL6, another key BTB/POZ family member[1]. ZBTB20 has garnered attention for its roles in tumorigenesis, being a hotspot for genetic variation and fusion in various cancers[1]. Pathogenic mutations in ZBTB20 are associated with Primrose syndrome and other developmental conditions[4]. No approved small-molecule drugs or biologics currently target ZBTB20 directly; however, its functional domains are considered promising therapeutic candidates in oncology and immune modulation[1][3].
