Overview

Zinc finger and BTB domain-containing protein 22 (ZBTB22) is a transcription factor characterized by an N-terminal BTB/POZ domain and C-terminal C2H2-type zinc finger motifs, which enable dimerization and sequence-specific DNA binding[4][2][3]. ZBTB22 directly regulates gene expression by binding GC-rich promoter regions and acts as a positive modulator of gluconeogenic and lipogenic genes in hepatocytes. Overexpression of ZBTB22 enhances hepatic glucose production by activating transcription of phosphoenolpyruvate carboxykinase (PCK1), contributing to hyperglycemia and insulin resistance in model systems[2]. Knockdown or knockout of ZBTB22 leads to decreased expression of gluconeogenic/lipogenic genes and increased fatty acid oxidation, implicating a role in metabolic reprogramming in liver and potentially in pathologies such as Type 2 diabetes and non-alcoholic fatty liver disease[2]. As a member of the POK (POZ and Krüppel) family of transcription factors, ZBTB22 is implicated broadly in the regulation of proliferation, differentiation, and development, although specific roles outside hepatic metabolism remain to be fully elucidated. No direct drugs or clinical biomarkers for ZBTB22 are reported to date, but its regulatory role in metabolism marks it as a putative therapeutic target in diabetes and metabolic syndrome[2][4].
