Overview

Zinc finger and BTB domain-containing protein 6 (ZBTB6) is a member of the ZBTB family, characterized by a C-terminal zinc finger domain enabling sequence-specific DNA binding and an N-terminal BTB (POZ) domain mediating protein–protein interactions[1][3]. It mainly acts as a transcriptional repressor, recruiting corepressors (such as N-CoR, SMRT, and HDACs) to gene promoters via its BTB domain. ZBTB6 is implicated in regulating negative transcription by RNA polymerase II, cytokine production, immune process regulation, cell cycle progression, apoptosis, and cell proliferation. It is highly expressed in several cancers, especially breast cancer, where it promotes tumor progression through repression of ARHGAP6 and activation of the STAT3 signaling pathway[1]. Its overexpression correlates with higher tumor grade and worse prognosis. ZBTB6 is considered a potential prognostic indicator and possible chemotherapeutic target in cancer[1][3][4].
