Overview

Zinc finger and SCAN domain-containing protein 1 (ZSCAN1) is a transcription factor characterized by a C2H2 zinc finger DNA-binding motif and a SCAN domain responsible for protein-protein interactions. It is encoded by the ZSCAN1 gene and is expressed in tissues such as the brain, pituitary gland, and testis, where it localizes to the nucleoli. ZSCAN1 regulates gene expression by binding to specific DNA sequences and may play roles in epigenetic regulation, stemness, and genomic integrity. Emerging evidence implicates ZSCAN1 as a tumor suppressor in breast cancer, where its downregulation predicts poor prognosis and its overexpression impairs cancer cell proliferation and stemness. ZSCAN1 autoantibodies have been detected in patients with paraneoplastic neurologic syndromes and certain tumors, highlighting its role in autoimmunity. Although ZSCAN1 is a biologically and clinically relevant protein, no drugs currently target it directly, nor is it widely established as a therapeutic target outside the context of autoimmunity and cancer prognosis.
