Overview

Zinc finger BED domain-containing protein 3 (ZBED3) is a member of the ZBED family characterized by a zinc finger BED domain. It is primarily located in the cytoplasm, where it acts as a positive regulator of the canonical Wnt/β-catenin signaling pathway by directly interacting with Axin, thereby stabilizing cytoplasmic β-catenin and promoting transcription of Wnt target genes[3][5]. ZBED3 is involved in cell proliferation, cancer progression, metabolic regulation (including insulin resistance and diabetes), stem cell differentiation, and oocyte maturation. Loss of ZBED3 in model organisms leads to impaired cytoskeletal and organelle dynamics in early embryogenesis and reduced fertility[1][2][5]. Although it is not currently the direct target of therapeutic drugs, its central role in cell signaling and disease makes it a topic of significant research interest.

Mechanism of action

Disrupts recruitment of kinases required for β-catenin degradation by interacting with Axin; stabilizes β-catenin in cytoplasm, leading to canonical Wnt pathway activation. Phosphorylation-dependent interaction via PPPPSPT motif.
