Overview

Zinc finger C3HC-type protein 1 (ZC3HC1, NIPA) is an F-box and zinc finger domain–containing nuclear protein encoded on chromosome 7q32.2. As part of the SCF-type E3 ubiquitin ligase complex (SCFNIPA), it regulates cell cycle progression by targeting cyclin B1 for degradation, thereby controlling entry into mitosis. ZC3HC1 expression and function are tightly regulated by cell cycle–dependent phosphorylation. It is broadly expressed in human tissues, especially the heart, skeletal muscle, and testis. Clinical relevance includes its association with coronary artery disease risk via the rs11556924 SNP, independent of traditional risk factors, making it a genetic biomarker for cardiovascular events. Additional associations exist with immunodeficiency and rare developmental syndromes[1][3][4][5][8]. No direct drug interactions or established safety concerns have been documented to date.
