Zinc finger CCCH-type containing 4 (ZC3H4) is a nucleic acid-binding protein characterized by a CCCH-type zinc finger motif, coordinating three cysteine and one histidine residues. ZC3H4 plays an essential role in suppressing widespread transcription of non-coding RNAs in human cells—functioning as part of transcription termination checkpoints in partnership with other factors, such as WDR82 and CK2, within the ZWC complex. By promoting the termination and exosome-mediated degradation of lncRNAs, ZC3H4 controls enhancer activity and maintains proper transcriptional directionality at gene promoters, restricting extragenic and antisense transcription. During early embryogenesis, ZC3H4 is required for successful lineage specification and blastocyst development, with loss-of-function mutations resulting in developmental failure, severe DNA breaks, and impaired cell proliferation. Currently, no drugs or therapeutic agents target ZC3H4, and it is not a receptor, enzyme, or transporter in human physiology.
