Overview

Zinc finger CCHC domain-containing protein 12 (ZCCHC12) is a transcriptional coactivator characterized by a CCHC-type zinc finger domain, primarily localizing to the nucleus.[4][6] Its principal function is to enhance bone morphogenetic protein (BMP) signaling by interacting with SMAD1 and CBP within transcriptional complexes, thereby influencing the expression of BMP-responsive genes, particularly in cholinergic neurons.[5][4][2] The protein is implicated in neurodevelopment, with genetic variations linked to X-linked intellectual disability and potentially other neurodevelopmental disorders.[4] ZCCHC12 is also identified as a metastasis-related oncogene in papillary thyroid cancer[1] and has been proposed as a marker for Cajal–Retzius cells and endometrial receptivity.[2] It does not function as a classic therapeutic target, receptor, or enzyme, and there are no approved drugs that directly act on ZCCHC12.
