Overview

Zinc finger CCHC domain-containing protein 17 (ZCCHC17) is a highly conserved nucleolar protein characterized by an S1 RNA-binding region and a CCHC-type zinc finger motif; it is predominantly expressed in the nucleolus where it is involved in ribosome maturation and biogenesis. ZCCHC17 interacts with splicing factors and is a negative regulator of rRNA transcription via interference with upstream binding factor (UBF) on rDNA promoters, leading to reduced pre-rRNA synthesis and nucleolar stress when overexpressed. In neurons, ZCCHC17 plays a critical role in regulating synaptic gene expression and maintaining synaptic protein balance; its reduced expression correlates with cognitive decline and tau pathology in Alzheimer's disease, making it a novel therapeutic target for enhancing cognitive resilience. The protein is a core component of the 60S ribosomal subunit, impacts mRNA quality control, and may regulate splicing and export of poly(A)+ RNA in the nucleus.
