Overview

Zinc finger CCHC domain-containing protein 3 (ZCCHC3) is a member of the zinc-knuckle (CCHC-type zinc finger) protein superfamily, characterized by motifs that specifically bind single-stranded nucleic acids, especially RNA. ZCCHC3 functions as an RNA-binding protein involved in the innate immune response to viral infection, facilitating detection and restriction of viral and retrotransposon (e.g., LINE-1) RNAs. It interacts with viral nucleic acids, aids in the formation of stress granules under cell stress, and associates with the RNA exosome, supporting RNA surveillance and turnover. ZCCHC3 helps cells defend against retrovirus propagation and transposon mobility, and it has been shown to physically interact with proteins and RNA complexes linked to antiviral defense. While ZCCHC3 is strongly implicated in antiviral immunity and RNA processing, it is not a current therapeutic target (not a conventional receptor, enzyme, transporter, or direct drug target). The zinc-knuckle domain is central for its RNA-binding and regulatory functions. No drugs or clinical inhibitors targeting ZCCHC3 are known. Its roles in disease are mostly indirect, by restricting the mobility of elements that could otherwise contribute to genomic instability or infection. It does not fit the standard classes of druggable targets such as GPCRs, ion channels, or enzymes. No evidence for biomarker or safety issue reporting in the scientific literature.
