Zinc finger CCHC-type and RNA-binding motif-containing protein 1 (ZCRB1) is a highly conserved RNA-binding protein that serves as a core component of the U12-type (minor) spliceosome, specifically within the U11/U12 small nuclear ribonucleoprotein (snRNP) complex. It is essential for the correct splicing of minor intron-containing genes (MIGs) and, by extension, for proper expression of genes involved in processes such as ciliogenesis, cell signaling, and tissue morphogenesis. Partial or complete loss of ZCRB1 results in impaired minor intron splicing, dysregulation of cilia-related genes, and defective primary cilia formation, as well as upregulation of WNT signaling pathways. These disruptions may lead to developmental abnormalities and are linked to essential cellular homeostasis and survival. The protein's role is not restricted to a single tissue or process, as its function in minor splicing is fundamental and broadly required for cell viability in vertebrates.
