Overview

Zinc finger CW-type and PWWP domain-containing protein 2 (ZCWPW2) is a nuclear protein expressed in humans that functions as a "histone methylation reader," specifically recognizing lysine 4 methylation states (H3K4me0, H3K4me1, H3K4me2, H3K4me3) on histone H3[6][9]. It contains highly conserved CW-type zinc finger and PWWP domains, structural motifs that allow it to bind specific histone modifications and contribute to chromatin structure regulation[4]. ZCWPW2 is tightly co-evolved with the methyltransferase PRDM9 and is hypothesized to localize to sites of PRDM9 binding; this suggests it may participate in meiotic recombination, possibly by recruiting or positioning recombination machinery during double-strand break (DSB) formation or repair[2]. The detailed molecular function of ZCWPW2 remains uncertain, but, like related zinc finger proteins, it is likely involved in transcriptional regulation, chromatin remodeling, and maintaining genome integrity. There are no established drugs targeting ZCWPW2, nor is it considered a classical therapeutic target, and no direct disease associations have been defined[2][6][9].
