Zinc finger DHHC domain-containing protein 11 (ZDHHC11) is a member of the DHHC family of palmitoyltransferases localized to the endoplasmic reticulum. It catalyzes the palmitoylation of protein substrates, notably NCDN, and regulates membrane targeting and protein-protein interactions via this modification. ZDHHC11 also acts as a signaling adaptor that modulates innate immune responses, functioning independently of its catalytic activity to enhance recruitment of IRF3 to STING1, thereby promoting activation of type I interferon pathways in response to DNA viruses. Importantly, ZDHHC11 facilitates NF-κB activation by binding and promoting oligomerization of TRAF6, boosting the E3 ubiquitin ligase activity essential for inflammatory signaling and cytokine production such as IL-8 and IL-6. ZDHHC11 is therefore emerging as a regulatory element in inflammation, antiviral defense, and potentially cancer biology through its influence on NF-κB signaling.
