Zinc finger DHHC-type containing protein 24 (ZDHHC24) is an enzyme that catalyzes the S-palmitoylation of proteins, a post-translational modification where palmitate is covalently attached to cysteine residues on substrate proteins[2]. ZDHHC24 is part of the large ZDHHC family of palmitoyltransferases that regulate protein trafficking, membrane localization, and stability. Its expression and function affect diverse cellular processes, and dysregulation is linked to cancer, especially lung adenocarcinoma, where ZDHHC24 serves as a poor prognostic marker and correlates with immune microenvironment alterations[1]. ZDHHC24 is also implicated in Bardet-Biedl syndrome[2]. There are currently no approved drugs targeting ZDHHC24 specifically, and existing inhibitors lack isoform selectivity, presenting both safety and efficacy challenges for therapeutic development targeting this enzyme[3][4].
