Overview

Zinc finger E-box-binding homeobox 2 (ZEB2) is a transcription factor containing multiple zinc finger domains and a homeodomain, encoded by the ZEB2 gene. ZEB2 acts primarily as a transcriptional repressor by binding to E-box sequences in DNA and interacting with corepressor complexes such as NuRD and CtBP. It regulates cell fate decisions, differentiation (notably in the nervous system, hematopoietic and immune cells), and is a key effector of TGF-β signaling, essential for embryogenesis and tissue homeostasis. Germline mutations in ZEB2 cause Mowat–Wilson syndrome, and its dysregulation contributes to developmental defects and cancer progression, often by promoting epithelial–mesenchymal transition and stem-like features in tumor cells.
