Overview

Zinc finger FYVE domain-containing protein 16 (ZFYVE16 or Endofin) is a human protein encoded by the ZFYVE16 gene. It contains a FYVE-type zinc finger domain that binds phosphatidylinositol 3-phosphate, facilitating its localization to endosomal membranes. ZFYVE16 functions as a scaffold protein that brings together the TGF-β type I receptor and Smad4 to enable efficient TGF-β/Smad signal transduction, as well as modulating bone morphogenetic protein (BMP) pathway activity. ZFYVE16 is also implicated in the modulation of the amplitude of MAPK signaling upon tyrosine phosphorylation. It interacts with other endosomal proteins such as TOM1, thus helping to regulate membrane trafficking and endosome function. Although it plays a central role in several cellular signal transduction cascades and intracellular transport, ZFYVE16 is not considered a classical therapeutic target, and no drugs are known to act on it directly.
