Overview

Zinc finger FYVE domain-containing protein 19 (ZFYVE19) is a metal binding protein that plays a critical role in the regulation of the abscission step in cytokinesis, specifically as part of the NoCut checkpoint pathway, which delays cell separation to ensure proper chromosome segregation. The protein localizes primarily at the centrosome and binds phosphatidylinositol-3-phosphate, indicating involvement in lipid and protein interactions during cell division. Pathogenic mutations, particularly biallelic loss-of-function variants, in ZFYVE19 are linked to pediatric-onset cholestatic liver diseases, including congenital hepatic fibrosis, sclerosing cholangiopathy, and high-GGT cholestasis. There is currently no evidence supporting direct targeting of ZFYVE19 by therapeutic drugs, nor is it commonly used as a biomarker or therapeutic target in clinical settings.
