Overview

Zinc finger FYVE domain-containing protein 26 (ZFYVE26), also known as spastizin, is a multifunctional adaptor protein critical for membrane trafficking, especially at the midbody during cell division and in autophagy. Its FYVE zinc finger domain allows binding to phosphatidylinositol 3-phosphate (PI3P), targeting ZFYVE26 to distinct membrane compartments involved in cytokinesis and intracellular transport. ZFYVE26 is essential for the maturation and clearance of autophagosomes and thereby for the maintenance of lysosomal and neuronal homeostasis. Mutations in ZFYVE26 cause autosomal recessive spastic paraplegia type 15 (SPG15), a progressive neurodegenerative disorder marked by lower limb spasticity, cognitive decline, and neuroimaging findings such as thinning of the corpus callosum. ZFYVE26 interacts with a range of endosomal and autophagy factors, including components of the ESCRT-III and AP-5 complexes, but is not currently considered a therapeutic target for approved pharmaceuticals[1][2][3][4][5].
