Overview

Zinc finger FYVE domain-containing protein 9 (ZFYVE9) is an early endosomal adapter protein known for its role as a scaffold in the transforming growth factor-beta (TGF-beta) signaling pathway. It functions by recruiting SMAD2/3 proteins to the TGF-beta receptor complex at the endosomal membrane, facilitating their phosphorylation and subsequent nuclear translocation, which is essential for TGF-beta–mediated transcriptional responses. ZFYVE9/SARA contains a FYVE-type zinc finger domain that targets it to endosomal membranes by recognizing phosphatidylinositol 3-phosphate. It influences several cellular processes through SMAD signaling, including cell growth, differentiation, and development. Dysregulation of ZFYVE9 or its pathway partners can contribute to certain pathologies, such as cancer and immune-mediated glomerulonephritis[1][3][6][9][8].
