Zinc finger FYVE-type containing 28 is a human protein encoded by the ZFYVE28 gene, localized primarily in the cytosol and early endosome membrane[1][2][3][4][5]. It contains a FYVE domain which mediates phosphatidylinositol-3-phosphate binding. ZFYVE28 acts as a negative regulator of epidermal growth factor receptor (EGFR) signaling by promoting EGFR degradation in endosomes, particularly when the protein is not monoubiquitinated[1][5]. Additionally, ZFYVE28 has been reported to mediate insulin resistance by promoting the degradation of the phosphorylated insulin receptor via increased late endosome production[4]. Disease associations include certain rare autoimmune and infectious conditions, and it is involved in cellular pathways associated with cancer and metabolic disease[1][4].
