Zinc finger homeobox protein 3 (ZFHX3)

Target
ZFHX3
Molecular classification
Transcription factor, Zinc finger protein, Homeobox protein
01

Overview

Zinc finger homeobox protein 3 (ZFHX3) is a large transcription factor characterized by multiple zinc finger motifs and homeodomains. It regulates gene expression by binding to AT-rich motifs in enhancers and promoters, orchestrating diverse biological processes such as neuronal and myogenic differentiation, cell cycle regulation, and suppression of oncogenes. ZFHX3 has key physiological roles in tumor suppression, circadian rhythm regulation (notably in the suprachiasmatic nucleus), and cardiovascular function. Loss or mutation of ZFHX3 is associated with several cancers, cardiovascular diseases (especially atrial fibrillation), neurodevelopmental disorders, and inflammation. ZFHX3 can serve as a biomarker for certain cancer immunotherapies and for prognosis in atrial fibrillation and other disease states[1][2][3][4].

Mechanism of action

Transcriptional regulation by DNA binding at AT-rich enhancers and promoters[2][1]\nModulation of gene expression related to cell cycle, apoptosis, and differentiation[2]\nSuppression or activation of specific growth-control genes (e.g., alpha-fetoprotein, c-Myb, CDKN1A)[2]

Biological functions

Transcription regulationCell cycle regulationCell differentiation (neuronal, myogenic)Tumor suppressionCircadian rhythm regulationNeuronal survivalSuppression of oncogenes (e.g., c-Myb)Activation of cell cycle inhibitors (e.g., CDKN1A/p21CIP1)
04

Disease associations

Cancer (tumor suppressor in prostate, breast, gastric, endometrial, non-small cell lung cancers)Cardiovascular disease (predisposes to atrial fibrillation, implicated in heart failure, pediatric vasculitis)Neurodevelopmental disorders (intellectual disability, spinocerebellar ataxia)Inflammatory disorders
05

Safety considerations

No specific targeted small molecule drugs known; as a transcription factor and tumor suppressor, potential concerns would include broad effects on cell proliferation, differentiation, tumorigenesis, and adverse effects relating to systemic gene regulation if therapeutically targeted[2]. No direct safety data as a drug target was identified.
06

Biomarkers

ZFHX3 status (expression level/genetic variants) as a predictive biomarker for immune checkpoint inhibitor response in non-small cell lung cancer[2].Genetic variants (e.g., single nucleotide polymorphisms) associated with atrial fibrillation risk and recurrence[2].
