Zinc finger MYM-type protein 2 is a nuclear protein encoded by the ZMYM2 gene, characterized by multiple MYM-type zinc finger domains. It acts mainly as a transcriptional repressor and chromatin remodeler via interaction with multiprotein complexes like histone deacetylase assemblies. ZMYM2 plays roles in cell cycle progression, gene silencing, and embryonic development. Chromosomal translocations involving ZMYM2—most notably the ZMYM2-FGFR1 fusion—result in constitutively active kinase signaling, driving hematologic malignancies such as myeloproliferative neoplasms, lymphoma, and acute myeloid leukemia. ZMYM2 is also crucial for the proper regulation of DNA damage responses and for establishing repressive epigenetic states during development.
For fusion proteins (ZMYM2-FGFR1): drugs target aberrant tyrosine kinase signaling stemming from the fusion event, inhibiting downstream pathways (STAT5, AKT, MAPK). Histone deacetylase inhibitors could theoretically alter ZMYM2-mediated gene silencing and chromatin remodeling.
