Zinc finger protein 213 (ZNF213) is a human transcription factor belonging to the C2H2 zinc finger protein family, containing KRAB and SCAN domains. ZNF213 functions in both the cytosol and nucleus to regulate gene expression and modulate protein-protein interactions. In breast cancer, ZNF213 acts as a key modulator of estrogen receptor alpha signaling by interacting directly with ERα to inhibit its ubiquitination and degradation, thereby enhancing ERα stability and promoting tumor cell proliferation. High expression of ZNF213 is associated with poor outcomes in patients receiving endocrine therapy, suggesting that it plays a role in therapy resistance and is a potential therapeutic target in ERα-positive breast cancer.
ZNF213 inhibition increases ERα protein ubiquitination and degradation, decreasing ERα signaling and cellular proliferation. Knockdown of ZNF213 sensitizes breast cancer cells to tamoxifen.
